Ku Shen (Sophora flavescens root) and Shan Dou Gen (Sophora tonkinensis root and rhizome) are closely related species within the Fabaceae family but differ in their chemical composition and traditional use contexts. Sophora flavescens is the species officially recognized in the Chinese Pharmacopoeia as the botanical source of Ku Shen (Sophorae Flavescentis Radix), while Sophora tonkinensis is the botanical source of Shan Dou Gen (Sophorae Tonkinensis Radix et Rhizoma). Both contain quinolizidine alkaloids including matrine and oxymatrine, but Ku Shen typically contains higher levels of these constituents. In TCM, Ku Shen is more strongly associated with killing parasites and stopping itching for skin conditions, while Shan Dou Gen has a stronger focus on clearing Heat and resolving toxicity for sore throat and mouth ulcers. While they share some overlapping constituents and uses, they are considered distinct herbs in the TCM tradition and are not interchangeable without careful consideration.
